Simultaneous quantification of TB-500 and its metabolites in in-vitro experiments and rats by UHPLC-Q-Exactive orbitrap MS/MS and their screening by wound healing activities in-vitro
Authors: Khandoker Asiqur Rahaman, Anca Raluca Muresan, Hophil Min, Junghyun Son, Hyung-Seop Han, Min-Jung Kang, Oh-Seung Kwon

Researchers validated a UHPLC-Q-Exactive Orbitrap MS method for TB-500 and its metabolites in human serum, in-vitro enzyme systems and rat urine. Ac-LK was the main metabolite in rats and Ac-LKK a long-term metabolite. In fibroblast scratch assays, only the metabolite Ac-LKKTE differed significantly from control, and no cytotoxicity was observed.
Investigation of in vitro/ex vivo TB-500 metabolism, synthesis of relevant metabolites and detection limits in urine and plasma
Authors: P. Van Eenoo

Using human liver microsomes, S9 fraction and plasma/serum, this study showed that TB-500 undergoes stepwise C-terminal cleavage while its N-terminal acetylation is protective. Three metabolites and a deuterated internal standard were synthesized, and detection limits of 50–500 pg/mL were established for urine screening.
Utilizing Developmentally Essential Secreted Peptides Such as Thymosin Beta-4 to Remind the Adult Organs of Their Embryonic State—New Directions in Anti-Aging Regenerative Therapies
